Caspases play a major role in the transduction of the apoptotic signal and execution of apoptosis in mammalian cells. Caspase 3 (CPP32) and Caspase 6 (Mch2) are the major active caspases in apoptotic cells, and are activated in response to distinct apoptosis inducing stimuli and in all cell lines analyzed. Both CPP32 and Mch2 are present in apoptotic cells as multiple active species. Caspase 6 cleaves nuclear mitotic apparatus protein (NuMA) and mediates the shrinkage and fragmentation of nuclei. Cellular Localization: Cytoplasmic. Positive Control: Jurkat cells. Applications: Suitable for use in Immunoprecipitation. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
