Caspase-9 is a member of the CED-3 subfamily, bearing high similarity to caspase-3. Procaspase-9 contains a long N-terminal putative prodomain with high similarity to the prodomains of CED-3 and caspase-2. It contains two potential processing sites between its large and small subunits. It was demonstrated that both caspase-3 and granzyme B activated pro-caspase-9, although to differently sized products. By cleavage of caspase-3, it generates two products of molecular masses C37 kD (p37) and C10 kD (p10). The ability of caspase-3 to activate pro-caspase-9 suggests that the latter is downstream of caspase-3 and, once activated, caspase-9 can initiate a caspase cascade involving the downstream executioner caspases-3, -6, and -7. The involvement of caspase-9 in the process of apoptosis in zebrafish model was also investigated. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA: 1:5000-1:20,000 Western Blot: 1:500-1:1000 Optimal dilutions to be determined by the researcher. Control Peptide: C2088-06N Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
