Caspase family proteases play an important role in apoptosis. Currently, the family consists of more than 10 members. Based on their extended substrate specificities, these enzymes were divided into three major groups:
Group I (contains caspase-1,-4,-5)
Group II (contains caspase-2,-3,-7)
Group III (contains caspase-6,-8,-9,-10).
Evidences have suggested that Group I caspases are involved primarily in the production of inflammatory cytokines, while group II and III enzymes function in apoptosis as effectors and upstream activators, respectively. Active recombinant human caspases were expressed in E. coli and routinely tested for their ability to enzymatically cleave the substrate VEID-pNA (for caspase-6), IETD-pNA (for caspase-8 & -10) and LEHD-pNA (for caspase-9), respectively.
Highly active recombinant human caspases were expressed in E. coli and purified. Active recombinant caspases are useful in screening caspase inhibitors, studying enzyme kinetics and regulation, determining target substrates and as a positive control in caspase activity assays. We recommend using 1 unit per assay for analyzing caspase activity.

UNIT DEFINITION:
One unit of the active recombinant caspases is the enzyme activity that cleaves 1nmol of the individual caspase substrate (YVAD-pNA for caspase-1, VDVAD-pNA for caspase-2, DEVD-pNA for caspase-3 & -7, WEHD-pNA for caspase-4 & caspase-5, VEID-pNA for caspase-6, IETD-pNA for caspase-8 & -10, and LEHD-pNA for caspase-9), per hour at 37 degrees C in a reaction solution containing 50mM Hepes, pH 7.2, 50mM NaCl, 0.1% Chaps, 10mM EDTA, 5% glycerol, and 10mM DTT.
Storage and Stability:
Reconstitute to nominal volume by adding sterile PBS, aliquot and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer
