CCL19, Human, BioAssay(TM) Kit (MIP-3 beta, ELC, C-C Motif Chemokine 19, MIP-3beta, ELC, EBI1Ligand Chemokine)

C-C motif chemokine 19 (CCL19), also known as MIP-3beta and ELC (EBI1Ligand Chemokine), is one of the small cytokines belonging to the CC chemokine family. CCL19 cDNA encodes a 98 amino acid (aa) residue precursor protein with a predicted 21aa residue signal peptide that is cleaved to form the 77aa residue mature secreted protein. CCL19 is distantly related to other beta chemokines (20-30% aa sequence identity) and the gene for CCL19/MIP-3beta has been mapped to chromosome 9p13 rather than chromosome 17 where the genes for many human beta chemokines are clustered. CCL19 has been shown to be constitutively expressed in various lymphoid tissues (including thymus, lymph nodes, appendix and spleen). The expression of CCL19 is downregulated by the anti-inflammatory cytokine IL-10. Recombinant CCL19 has been shown to be chemotactic for cultured human lymphocytes. CCL19 has also been shown to be a unique functional ligand for CCR7, a chemokine receptor expressed in various lymphoid tissues and activated B and T lymphocytes.\n\nIntended Use:\nDesigned to qualitatively and quantitatively determine the presence of cells expressing CCL19 receptors or other interacting proteins by flow cytometry.\n\nTest Principle:\nWashed cells are incubated with the biotinylated protein that in turn binds to the cells via specific cell surface receptors or other interacting proteins. The cells are then directly incubated with avidin-fluorescein, which attaches to the bound biotinylated protein. Cells expressing the receptor/interacting protein are fluorescently stained, with the intensity of staining proportional to the density of such protein.
