CD115, a cell membrane associated antigen which is also known as c-fms, Macrophage Colony Stimulating Factor Receptor (M-CSFR) and CSF-1R. Two glycoproteins are recognized, with Mr=130kD and 150kD, corresponding to the immature and mature forms of CD115, respectively. The immature form (gp130c-fms) and the mature form (gp150c-fms) differ in composition of N-linked oligosaccharide chains. CD115 is an integral membrane protein with a 512 amino acid CSF-1 binding portion, a 25 amino acid membrane spanning portion and a 435 amino acid cytoplasmic portion. The cytoplasmic portion contains a tyrosine protein kinase domain. This monoclonal antibody inhibits CSF-1 binding and ligand-dependent proliferation. It also interferes with CSF-1 independent growth observed in cells expressing a mutant oncogenic form of CSF-1R. It does not induce internalization or turnover of CSF-1R. Applications: Suitable for use in Immunohistochemistry and Immunofluorescence. Also suitable for use in blocking studies after removal of sodium azide. Other applications have not been tested. Recommended Dilution: Immunohistochemistry (acetone fixed cryostat sections only): 1:25 with a 30-60 minute incubation at 4 degrees C is recommended. Optimal dilutions to be determined by the researcher. Recommended Positive Control: Tonsil, normal monocytes and macrophages and their committed bone marrow progenitors. Lymphocytes and granulocytes are not suitable for this application. Hybridoma: Rat splenocytes fused with rat myeloma Y3-Ag1.2.3 cells. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
