Human CD11c (aX integrin) complexes with CD18 (B2 integrin) to form the complement receptor type 4 (CR4) heterodimer which binds to fibrinogen and is involved with monocyte/granulocyte adhesion during inflammatory responses. CD11c expression is restricted to leukocytes mainly of myeloid lineage with highest expression on macrophages.
Applications:
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilution:
Flow Cytometry: 5x10e5 ficoll prepared human peripheral blood mononuclear cells per tube were washed and pre-incubated 5 min with 20ul of 250ug/ml human IgG (To block non specific binding) after which they were incubated 45 min on ice with 80ul of anti-CD11c antibody at 10ug/ml. Cells were washed twice and incubated with 50ul of Goat anti-Mouse IgG/FITC, after which they were washed three times, fixed and analyzed by FACS. A 15% sub population of cells stained positive with a mean shift of 1.01 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
