Human CD14 is strongly expressed on myelomonocyte lineage cells including monocytes, macrophages, and a subset of granulocytes. CD14 is a high affinity receptor for complexes of lipopolysaccharide and lipopolysaccharide binding protein. A soluble form of CD14 also binds to LPS and LBP. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5x10e5 ficoll prepared human peripheral blood mononuclear cells were washed and pre-incubated 5 min with 20microl human IgG at 250ug/ml (to block non specific binding) after which they were incubated 45 min on ice with 80ul of anti-CD14 antibody at 10ug/ml. Cells were washed twice and incubated with Goat anti-Mouse IgG/FITC, after which they were washed three times, fixed and analyzed by FACS. A net 4.8% sub population of the cells stained positive with a mean shift of 1.6 log10 fluorescent units when compared to a Mouse IgG2a negative control at a similar concentration. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
