CD154 is expressed predominantly on activated CD4+ T lymphocytes, and acts as a costimulatory molecule in B cell differentiation/proliferation, immunoglobulin switching and germinal centre formation, through interaction with the CD40 receptor on B cells, and cytokines secreted by activated T cells. Applications: Suitable for use in ELISA, Immunohistochemistry (paraffin), and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (paraffin): 2.5-5.0ug/ml ELISA (indirect); 0.5-2.0ug/ml. Western Blot: 0.1-0.2ug/ml Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20 degrees C. Reconstitute with sterile buffer or ddH2O. Aliquot and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
