CD16 is known as low affinity IgG receptor III (FcgRIII). It is expressed as two distinct (CD16a and CD16b) forms. CD16a (FcgRIIIA) is a 50-65 kD polypeptide-anchored transmembrane protein. It is expressed on the surface of NK cells, activated monocytes, macrophages, and placental trophoblasts in humans. CD16b (FcgRIIIB) is a 48 kD glycosylphosphatidylinositol (GPI)-anchored protein. It is over 95% homologous to CD16a in extracellular domain and is expressed specifically on neutrophils. CD16 binds aggregated IgG or IgG-antigen complex that functions in NK cell activation, phagocytosis, and antibody-dependent cell-mediated cytotoxicity (ADCC). The 3G8 antibody blocks neutrophil phagocytosis and stimulates NK cell proliferation. Applications: Suitable for use in Flow Cytometry, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 20ul per million cells in 100ul volume Immunohistochemistry: acetone-fixed frozen tissue Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
