CD178, otherwise known as Fas Ligand/ CD95L, a 40kDa type II transmembrane glycoprotein and member of the TNF/NGF superfamily, expressed by activated T cells and NK cells, which can be induced on a variety of cells by radiation, heat shock, chemotherapeutic agents and viral infection. CD178 acts as a key effector of cytotoxicity and in the regulation of immune responses. The binding of CD178 to its receptor CD95 (Fas), induces Fas-mediated apoptosis of target cells, and may be involved in the induction of peripheral tolerance and neutrophil chemotaxis. The binding of decoy receptor 3 (DcR3) to CD178 has been shown to inhibit CD178-mediated apoptosis. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:25-1:200; 10ul labels 10e6 cells in 100ul Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
