Expression of human CD25 (the alpha subunit of the IL-2 receptor complex) is dramatically upregulated on activated T cells, B cells, NK cells and macrophages (3). Antibodies to CD25 are being studied for potential use as an immundirected therapy to selectively attack (kill) CD25 expressing cells (4). Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: Cultured human CEM were stimulated overnight in the presence of PMA (10ng/ml) and ionomycin (1uM). 5x10e5 cells per tube were incubated 45 min on ice with 80ul of anti-CD25/R-PE at a dilution factor of 1:50 (5ug/ml). Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.24 log10 fluorescent units when compared to a Mouse IgG1/APC negative control at a similar concentration. Binding was blocked when cells were pre-incubated 10 min with 20ul of 0.5mg/ml anti-CD25 antibody. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer. APC is light sensitive.
