Human CD26 is a T cell activation antigen which can be associated with costimulation (1). The significance of the dipeptidyl peptidase IV activity remains unknown. Recent characterization of adenosine deaminase binding to CD26 suggests an important immunoregulatory role (2).
Applications:
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilution:
Flow Cytometry: Ficoll prepared human peripheral blood mononuclear cells were stimulated in culture 1 day with 10ng/ml PMA and 1uM Ionomicin. Cells were harvested, and 5x10e5 per tube were washed and pre-incubated ~5 min with 20ul of human IgG at 250ug/ml (to block non specific binding), after which they were incubated 45 min on ice with 80ul of anti-CD26 antibody at 10ug/ml. Cells were washed twice and incubated with Goat anti-Mouse IgG/FITC, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.31 log10 fluorescent units when compared to a Mouse IgG2b negative control at a similar concentration.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
