Integrin a10b1 is one of twelve integrin family adhesion receptors that share the b1 subunit. The non-covalent heterodimer of 160kD a11 and 130kD b1/CD29 type I transmembrane glycoprotein subunits is expressed mainly on chondrocytes within cartilage, but also infibrous connective tissues such as heart valves and ligaments. The a10 extracellular domain (ECD) contains an I (inserted) domain which includes the ligand binding site. The b1 ECD contains a vWFA domain, which participates in binding. Each subunit then has a transmembrane sequence and a short cytoplasmic tail . The dimer is folded when it is least active. Divalent cations and intracellular (inside-out ) signaling convert it to its most active, extended and open conformation. The 1100aa human a10 extracellular domain (ECD) shares 88-92% aa sequence identity with mouse, rat , canine and bovine a10, while the 708aa human b1 ECD shares 92-96% aa sequence identity with rat , bovine, mouse, and feline b1. A reported a10 splice variant lacking ECD aa974-1012 was not expressed on the cell surface. I domain-containing b1 integrins a1b1, a2b2, a10b1 and a11b1 all bind collagens; all but a11b1 also bind laminins. During cartilage differentiation, a10b1 is thought to be the main integrin binding type II and IX cartilage collagens. However, deletion of mouse a10 causes a mild phenotype including slightly shortened bones and narrowed hypertrophic zones, indicating that another collagen-binding integrin, likely a2b1, may compensate for a10b1 functions. Migration of melanoma cells has been noted to correlate with a10b1 expression.\n\nSource:\nChinese Hamster Ovary cell line, CHO-derived. Gln17-Gly523, with a C-terminal 6-His tag.\n\nMolecular Weight: 160-175kD & 125-150kD (SDA-PAGE, reducing conditions) \n\nBiological Activity:\nMeasured by its binding ability in a functional ELISA. When mouse Laminin is coated at 10ug/ml, Integrin a10b1 binds with an apparant Kd<5nM. \n\nEndotoxin: (same/less than)1EU/ug (LAL) \n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Reconstitute with 200ug/ml PBS. Aliquot and store at -20 degrees C. Reconstituted product is stable for 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
