The human CD3/T cell receptor (TcR) complex is made up of at least five CD3 proteins in association with proteins of the TcR. The TcR recognizes antigens in association with MHC molecules after which protein chains of the CD3 complex mediate activation signals triggered by TcR antigen binding. CD3 is expressed on greater than 95% of circulating human peripheral T cells. Recognizes the 20kD episilon chain of the CD3 molecule complex. Applications: Suitable for use in Western Blot, Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Paraffin Flow Cytometry: 5x10e5 cultured human HPB-MLT cells were washed and incubated 45 min on ice with 80ul of anti-CD3 antibody at 5ug/ml. Cells were washed twice and incubated with Goat anti-Mouse IgG/FITC, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 2.20 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
