Human CD41 is a member of major platelet transmembrane glycoprotein complex (CD41/CD61) which mediates platelet aggregation and adhesion by serving as the receptor for fibrinogen and von Willebrand factor, fibronectin and vitronectin. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5x10e5 ficoll prepared human peripheral blood mononuclear cells were washed and pre-incubated 5 min with 20ul of 250ug/ml human IgG (to block non specific binding) after which they were incubated 45 min on ice with 80ul of anti-CD41a antibody at 5ug/ml. Cells were washed twice and incubated Streptavidin/R-PE after which they were washed three times, fixed and analyzed by FACS. A net 6% sub population of cells stained positive with a mean shift of 2.78 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. Binding was blocked when cells were pre-incubated 10 min with 20ul of 0.5mg/ml anti-CD41a antibody. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
