CD43, a 135kD Type I transmembrane sialoglycoprotein, abundantly expressed on the surface of T-lymphocytes and hematopoietic cells. It is a member of sialomucin family and contains a heavily O-glycosylated extracellular domain, a transmembrane domain and an intracellular cytoplasmic domain. It exists in 2 isoforms; an 115kD form expressed on most T-cells and a 135kD form expressed in resting CD8+ cells. Natural ligands of CD43 are ICAM-1, Galectin-1 and MHC-1 suggesting a complex role of this protein in Leukocyte adhesion mediated by integrins, T-cell activation signaling cascade, anti-apoptotic and pro-apoptotic signaling and anti-adhesive properties based on interaction of each isoform with its ligand. Pathological role of CD43 is indicated in Wiskott-Aldrich syndrome. Applications: Suitable for use in Immunohistochemistry. Other applications not tested. Recommended Dilution:: Immunohistochemistry (formalin fixed paraffin embedded): 1:50-1:100 Immunohistochemistry: Frozen Optimal dilutions to be determined by the researcher. Positive Control: Tonsil Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
