Human CD45 is expressed on all cells of hematopoietic origin, except erythrocytes. CD45 is a transmembrane tyrosine phosphatase which can exist in at least nine different isoforms resulting from tissue- specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. CD45 plays an essential role in lymphocyte activation. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5x10e5 ficoll prepared human peripheral blood mononuclear cells were washed and pre-incubated ~10 min with 20ul human IgG at 250ug/ml (to block nonspecific binding), after which they were incubated 45 min on ice with 80ul of anti-CD45/APC at a 1:50 dilution (10ug/ml). Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 2.6 log10 fluorescent units when compared to a Mouse IgG2a/APC negative control at a similar concentration. Binding was blocked when cells were pre-incubated with 20ul of 0.5mg/ml anti-CD45 antibody. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer. APC is light sensitive.
