The monoclonal antibody is directed against the CD49e-antigen, molecular weights: 17, 130kD, (GP Ic or VLA-5 alpha-chain), which can form distinct complexes with either the CD29-antigen (GP IIa or VLA beta-chain), resulting in the VLA-5 (alpha-5, beta-1) complex which is expressed on human platelets. Platelet VLA-5 functions as a fibronectin receptor and platelet adhesion to fibronectin is inhibited by this antibody.
Applications:
Suitable for use in Indirect Immunofluorescence, Flow Cytometry and Fluorescent Microscopy. Other applications not tested.
Recommended Dilution:
Immunofluorescence: For studies on platelets, care should be taken to avoid activation of platelets during drawing of blood. Platelet activation markers are weakly expressed on platelets prepared from directly fixed normal blood. The expression is max. on thrombin (1U/ml) stimulated washed platelets. Fix cells or platelets with 1% paraformaldehyde, wash and adjust to ~108/ml for further incubations.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
