The replication licensing system acts to ensure that no section of the genome is replicated more than once in a single cell cycle. The origin recognition complex (ORC) and CDC6/CDC18 are needed on chromatin before the licensing reaction can take place. During the transition from a growth-arrested to a proliferative state, transcription of mammalian CDC6 is regulated by the cell cycle transcription factor (E2F protein). Immunoblots show that minichromosome maintenance (MCM) proteins are not associated with chromatin. Competence of G1 phase nuclei to replicate in vitro coincides with maximum CDC6 accumulation and MCM protein binding to chromatin in vivo. Antibodies against CDC6 and MCM5 stain abnormal (neoplastically transformed) cells in cervical smears and sections with remarkably high specificity and sensitivity. Cellular Localization: Nuclear. Positive Control: Raji or MDA231 cells. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
