Chk1 is a serine/threonine-protein kinase involved in cell cycle arrest when DNA damage has occurred, and it plays an important role in DNA damage checkpoint, embryonic development and tumor suppression. Chk1 is activated in response to replication blocks and genotoxic stress, via phosphorylation of serines 317 and 345 (2).Activated Chk1 can inactivate cdc25c through phosphorylation at Serine 216. Phosphorylation of cdc25c prevents activation of the CDC2-cyclin B complex and prevents entry into M-phase. Applications: Suitable for use in Western Blot and Immunocytochemistry/Immunofluorescence. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000-1:2000 Immunocytochemistry/Immunofluorescence: 1:100-1:250 Optimal dilutions to be determined by the researcher. Positive Controls: 283T cell lysate treated with UV and AP; HeLa cells Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
