Collagen IV antibody may be useful for the demonstration of basal lamina in invasive epithelial tumors. This antibody enables the discrimination between benign and malignant forms of soft tissue tumors (integrity of basement membranes is impaired in the latter case).\nThe extent of expression of the basement membrane component type IV collagen may be correlated with the degree of differentiation of basement membrane producing neoplasms. Neoplastic cells of invasive carcinomas often lack a continuous basement membrane. In diagnostic histopathology antibodies to type IV collagen may help to distinguish between non-invasive (benign or in situ) and invasive lesions. Furthermore the antibody is important in the differential diagnosis of bullous lesions in dermatopathology. In in vitro studies with endothelial cells and smooth muscle cells the antibody is helpful in detection of production collagen type IV.\n\nApplications: \nSuitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nImmunohistochemistry (frozen): 1:20; Exclusively stains basement membranes on cryostat sections of human tissues such as kidney, liver and heart. Stains canine Collagen IV on cryostat sections. Fixation with acetone for 5 min. at -20 degrees C is recommended.\nImmunohistochemistry (paraffin): 1:20; Stains canine Collagen IV on paraffin sections. Fixation with neutral formalin and proteolytic pretreatment with pepsin is recommended. Paraffin material generally renders less successful results.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
