Collagen Type VII

Type VII collagen is a newly characterized collagen which appears to be the major component of the anchoring fibril. \nType VII collagen fibrils are composed of three identical alpha collagen chains. It's location is restricted to the basement zone beneath stratified squamous epithelia. It functions as an anchoring fibril between the external epithelia and the underlying stroma. Mutations in the collagen V11 gene are associated with all forms of dystrophic epidermolysis bullosa.\n\nApplications: \nSuitable for use in Electron Microscopy, Immunofluorescence and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilution:\nImmunohistochemistry: Acetone-fixed frozen only. Fixation in methanol is not suitable.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nNormal skin\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes human intact (250kD) and collagenase digested (150kD) tissue Type VII collagen. Does not crossreact with types I, II, III, IV, V collagens, lamins or fibronectin. Epitope: Located on non-helical (non-collagenous) C-terminal region of collagen VII and is resistant to collagenase, trypsin, pronase, neuraminidase, chondroitinase, and hyaluronidase. It is, however, lost by digestion of collagen VII with pepsin. Species Crossreactivity: marmoset, monkey, bovine, sheep, goat, porcine and guinea pig.
