Type VII collagen fibrils are composed of three identical alpha collagen chains. It's location is restricted to the basement zone beneath stratified squamous epithelia. It functions as an anchoring fibril between the external epithelia and the underlying stroma. Mutations in the collagen V11 gene are associated with all forms of dystrophic epidermolysis bullosa. Applications: Suitable for use in Immunofluorescence, ELISA, Dot Blot, Western Blot, Immunohistochemistry, Immunocytochemistry and Immunoelectron Microscopy. Other applications not tested. Recommended Dilutions: Immunofluorescence (Indirect): 1:1000 on human or other mammalian frozen sections Immunohistochemistry (Frozen sections): 1:2000 on human tongue section fixed in acetone. Specifically stains basement membranes. Optimal dilutions to be determined by the researcher. Cellular Localization: Secreted, extracellular space, extracellular matrix, basement membrane Hybridoma: Fusion of mouse myeloma cells and splenocytes from BALB/c mice Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Additional info: Recognizes intact human tissue Type VII collagen (250kD). Pepsin digestion of terminal regions leads to complete loss of reactivity. Reactivity is conserved after treatment with collagenase. No reactivity against collagen types I, II, III, IV, V, laminin and fibronectin. Reacts predominantly with the basement membrane zone of stratified squamous epithelia and ectodermally derived glands. Immunoelectron microscopy shows antibody binding localized at the inferior border of the lamina densa. Species Crossreactivity: bovine, porcine, goat, guinea pig, monkey, marmoset and sheep.
