Collagen Type X (BSA & Azide Free)

Type X collagen is a non-fibrillar, short chain, network-forming collagen found in fetal hypertrophic cartilage in the growth zones of long bones, vertebrate, and ribs. It is a short collagen with a triple helical core half the length of the fibril-forming collagen type I, II and III and a globular domain at the carboxyl end (NC-1 domain) through which the molecules assemble into a hexagonal meshwork. The function of collagen X, a unique homotrimer synthesized by hypertrophic chondrocytes, is not known but its localization and transient expression at sites of calcification suggest that is is likely to be associated with events in the early stages of endochondral bone formation.\n\nApplications:\nSuitable for use in Immunofluorescence and Immunohistochemistry. Other applications have not been not tested.\n\nRecommended Dilution:\nImmunohistochemistry (frozen and formalin/paraffin): 2-4ug/ml for 30 min at RT.\nNote: Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min. Staining is further enhanced if heat induced epitope retrieval is followed by digestion of tissue sections with 0.05% hyaluronidase at 37 degrees C for 30 min.\n\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nChicken cartilage\n\nCellular Localization:\nCartilage matrix.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C or colder. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
