Collagen Type X

Type X collagen is a non-fibrillar, short-chain, network-forming collagen found in fetal hypertrophic cartilage in the growth zones of long bones, vertebrae, and ribs. It is a short collagen with a triple helical core half the length of the fibril-forming collagen type I, II, and III and a globular domain at the carboxyl end (NC-1 domain) through which the molecules assemble into an hexagonal meshwork. The function of collagen X, a unique homotrimer synthesized by hypertrophic chondrocytes, is not known but its localization and transient expression at sites of calcification suggest that it is likely to be associated with events in the early stages of endochondral bone formation. Endochondral ossification in growth plates proceeds through several consecutive steps of late cartilage differentiation leading to chondrocyte hypertrophy, vascular invasion, and, eventually, to replacement of the tissue by bone. By a combination of somatic cell hybrid screening and hybridization in situ the human collagen X gene (COL10A1) has been assigned to the distal end of the long arm of chromosome 6 at the locus 6q21-6q22.3. Osteoarthritis (OA) is a disorder characterized by new bone formation but the role of type X collagen in its pathogenesis is unclear. \n\nApplications: \nSuitable for use in Immunofluorescence and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilution:\nImmunohistology (frozen and formalin/paraffin): 2-4ug/ml for 30 minutes at RT.\nStaining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 minutes followed by cooling at room temperature for 20 minutes. Staining is further enhanced if heat-induced epitope retrieval is followed by digestion of tissue sections with 0.05% hyaluronidase at 37 degrees C for 30 minutes.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nChicken cartilage.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
