Human Complement component 3, C3 (alternative names include acylation-stimulating protein (ASP) C3 is encoded by gene located 19p13.3-p13.2. Because C3, C4, and C5 are strikingly similar suggesting a common evolutionary origin. C3 is an acute phase reactant. Synthesis of C3, a glycoprotein, is induced during acute inflammation. The liver is the main site of synthesis, although small amounts are also produced by activated monocytes and macrophages. A single chain precursor (pro-C3) of approximately 200kD is found intracellularly; the cDNA shows that it comprises 1,663aa. This is processed by proteolytic cleavage into alpha (~115kD) and beta subunits (~75kD) which in the mature protein are linked by disulfide bonds. Applications: Suitable for use in ELISA, Immunoelectrophoresis and single and double Radial Immunodiffusion (Mancini, Ouchterlony) to identify the presence of complement C3c or to determine its concentration. Other applications not tested. Recommended Dilution: immunoelectrophoresis: 2ul or equivalent against 120ul antiserum. Double radial immunodiffusion (Ouchterlony): Use a rosette arrangement with 10ul antiserum in a 3mm diameter centre well and 2ul serum samples (neat and serially diluted) in 2mm diameter peripheral wells. Single immunodiffusion: 1% antiserum in agar gel to establish a standard curve. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional info: Recognizes human Complement Factor C3c. May also react with the fragments C3b, C3bi and smaller fragments, since they all carry antigenic determinants of the C3c domain. Does not react with any other proteins component of human serum or plasma. In immunoelectrophoresis against fresh human serum, a single precipitin line is obtained in the beta-1 region representing native C3. Against serum containing partly activated C3, a precipitin line is obtained which extends from the beta-1 into the alpha-2 region, demonstrating a gradient. In old serum containing totally activated C3 a single precipitin line in the alpha-2 region is obtained.
