Complement Factor D

Factor D is a serine protease that catalyzes the initial proteolytic step in the alternative pathway of complement. Also known as adipsin, Factor D is expressed in adipose tissues at high levels.\n\nHybridoma:\nProduced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, NS0-derived, recombinant human Complement Factor D (rhFactor D; aa26-253: Accession # CAC48304). \n\nApplications:\nSuitable for use in ELISA (capture) and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nELISA Capture: This antibody can be used as a capture antibody in a human Complement Factor D ELISA in combination with biotinylated, affinity purified anti-human Complement Factor D polyclonal detection antibody (see catalog #C7850-70E). A general protocol is provided on the next page. Using plates coated with 100ul/well of the capture antibody at 4ug/ml, in combination with 100ul/well of the detection antibody, an ELISA for sample volumes of 100ul can be obtained. To arrive at the optimal dose range for this ELISA, set up a two-fold dilution series of the protein standard starting at 2ng/ml.\nWestern Blot: This antibody can be used at 1-2ug/ml with the appropriate secondary reagents to detect human Complement Factor D. Using a colorimetric detection system, the detection limit for rh Complement Factor D is ~10ng/lane under non-reducing conditions. Use of this antibody under reducing conditions is not recommended. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. In this application, the use of human Complement Factor D monoclonal antibody is recommended. \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
