Cortactin (p80,85)

Cortactin is present as a p80 and p85kD protein in many cell types. The protein was first identified as being a major substrate of the tyrosine kinase pp60SRC. The main structural feature of the protein is that at the N-terminus there are five and a half 37-amino acid repeats, these appear to be responsible for the actin-binding. This repeat region occupies about a third of the protein. The repeat region is followed by a helical region and then a proline rich region and finally a SH3 domain.\n\nApplications:\nSuitable for use in Western Blot, Immunoprecipation and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 0.5-2ug/ml detects Cortactin (p80/85) in mouse 3T3/A31 cell lysates. Detects Cortactin in chicken fibroblast src-transformed cell lysate. 3T3/A31 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C7901-80. Proteins were visualized using an IgG (HRP) Gt x Mo and a chemiluminescence detection system. \nImmunoprecipitation: Use 5ug for ~500ug of cell lysate.\nImmunocytochemistry: 10ug/ml\nOptimal dilutions to be determined by the researcher.\n\nHybridoma: \nSp2/0 myeloma x AJ mouse splenocytes\n\nIncluded Positive Antigen Control: \nF4209: 3T3/A31 cell lysate. Add 2.5ul 2-mercaptoethanol/100ul of lysate. Boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane.\nNote: PC-12 cell lysate, HeLa whole cell lysate or A431 whole cell lysate can also be used as a positive control.\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
