CrkL (Crk-like), a 36kD protein predominantly expressed in hematopoietic cell lines, is a member of the Crk adapter protein family. Lacking any intrinsic enzymatic activity, CrkL is composed of one SH2 domain and two SH3 domains (1). The SH2 domain of CrkL as been shown to interact with c-Cbl, Cas, Hef-1 and Paxillin (2); while the N-terminal SH3 can bind to c-ABL, Sos1 and C3G (3). Additionally, the phosphorylated form of CrkL (pY207) binds to BCr-ABL oncogene in chronic myelogenous leukemia cells (4).
Applications:
Suitable for use in Western Blot, Immunoprecipitation. Other applications not tested.
Recommended Dilution:
Western Blot: 1:10,000
Immunoprecipitation: 1:50
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
