Cutaneous Lymphocyte-assiciated Antigen (CLA) is a 140kD homodimer protein recognized by a unique monoclonal antibody. It is expressed on T lymphocytes in skin, subsets of peripheral blood memory T cells, NK cells, memory B cells and dendritic cells, as well as on monocytes, granulocytes and activated endothelial cells. CLA is a carbohydrate epitope of sialic acid- and fucose-modified P-selectin glycoprotein ligand-1 (PSGL-1), a surface glycoprotein expressed on majority of peripheral blood leukocytes. CLA is a subset ligand for E-selectin, P-selectin, and L-selectin. It plays a role in memory lymphocyte homing, tethering and rolling. Treatment of activated HUVEC cells with this antibody inhibits lymphocyte adhesion. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: (same/less than)2ug/10e5 cells in 100ul Immunohistochemistry: Acetone-fixed, frozen Immunohistochemistry: Formalin fixed paraffin embedded Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
