Cyclin A

In eukaroytic cells, the cell cycle is mediated by the sequential, activation and inactivation of complexes containing cyclin dependent kinases (CDKs) and cyclins. A number of CDKs have been identified and the activity of these various CDK family members is regulated by multiple mechanisms including binding to specific cyclin proteins, binding to specific small molecular weight inhibitory protein, and reversible phosphorylation. Binding of different cyclin protein is required to activate the catalytically inactive CDKs and determines the substrate specificity of the kinase. On the other hand, CDK inhibitors exert their inhibitory effect by blocking the catalytic activity of the cyclin-bound CDKs. Since the concentration of the CDKs are in excess and remain fairly constant throughout the cell cycle, their activation is determined by the availability of the different cyclin proteins and inhibitory factors. \n\nApplications: \nSuitable for use in Immunofluorescence, ELISA, Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilutions:\nImmunofluorescence: 5-10ug/ml detects Cyclin A in bovine, human, monkey and rat\nELISA: 0.1-1ug/ml \nWestern Blot: 1-2ug/ml detects Cyclin A in A431 cell lysate, bovine, HeLa cell lysate, human, monkey and mouse. Very sensitive for active Cyclin A protein.\nImmunoprecipitation: 2.5-5ug/ml detects Cyclin A in bovine, human, monkey and mouse\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Speci?c for the 60kD cyclin A protein and can be utilized to detect human, monkey, mouse and bovine cyclin A. Crossreactivity with other cyclin family members has not been detected.
