Cyclin D1,2

Cyclin D1 gene is identical with the BCL-1 and the PRAD-1 oncogene and is part of the chromosome 11q13 amplicon which is amplified in several common tumor types like breast, lung, esophagus, urinary bladder and head and neck carcinoma. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. About 50-70% of mantle cell lymphomas (MCL) contain a t (11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. Cyclins were originally identified as proteins whose levels fluctuated during the cell cycle. Subsequent studies have shown the cyclins control entry into different phases of the cycle cell by regulating the activity of cyclin-dependent kinases (CDKs). Mitotic or G2 cyclins regulate entry into M phase, whereas G1 cyclins regulate entry into S phase. Cyclin D1, a G1 cyclin, is essential for control at the G1/S transition (START in yeast), as it regulates the activity of CDK4 and CDK6 protein kinases. Translocation of the gene encoding cyclin D1, the bcl-1 oncogene, is associated with human B-lymphocytic malignancy, suggesting that alteration of cell cycle progression may be oncogenic.\n\nApplications: \nSuitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilutions:\nImmunoblot Analysis: 0.5-2ug/ml detects cyclin D1 and D2 in RIPA lysates from human A431 cells. Non-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C8454-10 (2ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.\nImmunoprecipitation: 5ug immunoprecipitates cyclin D1 and D2 from 500ug of A431 RIPA lysate. \nImmunohistochemistry:Reported to detect cyclin D1 and D2 in paraffin-embedded sections using high energy antigen retrieval (10mM citrate buffer, pH 6.0, microwaved 3-5 minutes, three times).\nOptimal dilutions to be determined by the researcher.\n\nPositive Antigen Control: A0001-50: A431 Cells, Non-Stimulated in RIPA Buffer, Sample Buffer\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes p36 cyclin D1 and crossreacts with p34 cyclin D2, which has a highly homologous epitope. No crossreactivity with cyclin A, cyclin B or cyclin D3. Species crossreactivity: human, mouse, rat and hamster.
