Cyclin D1 (PRAD-1, bcl 1)

Cyclins are regulatory subunits of the cyclin-dependent kinases (cdk), which control transition at different specific phases of the cell cycle. In mammalian cells, cyclin D1 plays a critical role in G1 to S transition. Cyclin D1 is a putative proto-oncogene overexpressed in a wide variety of human neoplasms.\n\nApplications: \nSuitable for use in Western Blot, Immunofluorescence, Immunoprecipitation, Immunohistochemistry and Flow Cytometry. Other applications not tested.\n\nRecommended Dilutions:\nImmunohistochemistry: 1:50-1:100 on frozen, formalin fixed or methacarn-fixed paraffin embedded sections. Staining of formalin-fixed paraffin-embedded tissue sections requires boiling in 10mM citrate buffer, pH 6.0 for 10-20 minutes followed by cooling at RT for 20 min prior to addition of antibody.\nImmunoprecipitation (Non-denatured and denatured): 10ul/mg protein lysate\nFlow Cytometry: 0.25ug stains 10e6 human PBMCs fixed in 100% methanol\nWestern Blot: 1:250-1:500\nOptimal dilutions to be determined by the researcher.\n\nHybridoma: \nNS2 myeloma cells with spleen cells from Balb/c mice. \n\nPositive Control: \nT47D, ZR75, BT474, SKBR3, MCF-7, MDA-MB-453, U-2-OS, HT29, Ramos, Jurkat, KNRK, C32 or A431. About 50-70% of mantle cell lymphomas and 40% of greast carcinomas are positive.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
