Cyclin E is a regulatory subunit of cdk-2 and is thought to be rate limiting for the G1/S transition during the mammalian cell cycle. Multiple isoforms of cyclin E are expressed only in tumors but not in the normal tissues, suggesting a post-transcriptional regulation of cyclin E. In vitro analyses indicated that these truncated variant isoforms of cyclin E are able to phosphorylate histone H1. Qualitative and quantitative alterations in cyclin E protein have been implicated as indicators of worse prognosis in various cancers. Ab-1 is superb for biochemical applications including co-precipitation of cdk2. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA: 1:2000 Western Blot: 1:200-1:500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
