Cyclin E1 is a member of the cyclin E family that can associate with and activate cyclin-dependent kinase Cdk2. Expression of cyclin E1 is essential for the control of the cell cycle at the late G1 and early S phase. The cyclin E/Cdk2 complex plays an essential and limiting role in the transition between G1 and S phase as well as the initiation of DNA replication. Ubiquination by the Cul-3 pathway and by Fbw7 regulates cyclin E1 levels and is critically important in normal cells. Abnormally high levels of cyclin E expression have frequently been observed in human cancers. Moreover, high cyclin E1 levels correlate with increased tumor aggression and poor prognosis.
