Cyr61, also known as IGFBP-10 and CCN1, is a 50kD matricellular glycoprotein that regulates the growth and adhesion of vascular endothelial cells, fibroblasts, and monocytes. Cyr61 induces VEGF upregulation, angiogenesis, and tumorigenesis through interactions with integrins alpha V beta 3, alpha V beta 5, alpha M beta 2, and alpha 6 beta 1. Cyr61 is cleaved by plasmin within its VWF domain which generates an N-terminal fragment that is not associated with the matrix but retains the ability to induce endothelial cell migration.\n\nIntended Use:\nFor the quantitative determination of human Cysteine-rich angiogenic inducer 61 (Cyr61) concentrations in cell culture supernates, serum, plasma, saliva, and human milk.\n\nSensitivity:\n3.8pg/ml\n\nRange:\n~39.1-2500pg/ml\n\nTest Principle:\nCyr61/CCN1, Human, BioAssay(TM) ELISA Kit employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for Cyr61 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Cyr61 present is bound by the immobilized antibody. After washing away any unbound substances, an enzyme-linked polyclonal antibody specific for Cyr61 is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Cyr61 bound in the initial step. The color development is stopped and the intensity of the color is measured.\n\nKit Components:\nMicroplate, 1x96wells\nStandard, 1x3vials \nConjugate (HRP), 1x21ml\nAssay Diluent, 1x11ml\nCalibrator Diluent, 1x21ml\nWash Buffer concentrate, 1x21ml\nColor Reagent A, 1x12ml\nColor Reagent B, 1x12ml\nStop Solution, 1x6ml\nPlate Sealers, 1x4 adhesive strips\n\nStorage and Stability:\nStore other components at 4 degrees C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
