Cytochrome C/Apaf-2 (apoptosis protease activating factor-2) is well known for its role in electron transport. During apoptosis (programmed cell death), Cytochrome C is released from the mitochondria into the cytosol where it binds to Apaf-1. Apaf-1 then activates caspase 9. Caspase 9 activation triggers other downstream caspase events in the proteolytic cascade. The mechanism by which Cytochrome C is released from the mitochondria and its function in Caspase 9 activation is still unclear. Applications: Suitable for use in Western Blot. Other applications have not been tested. Recommended Dilution: Western Blot: 1ug/ml Cell extracts from 293T cells (2x10e6) were separated into cytoplasmic and nuclear fraction, resolved by SDS-PAGE under reducing conditions, transferred to nitrocellulose and probed with C9095-09A at 1ug/ml. Proteins were visualized using a peroxidase-conjugated antibody to mouse IgG and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher. *Recommended Secondary Antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
