Cytokeratin 19

Cytokeratins are a subfamily of intermediate filament proteins and are characterized by a remarkable biochemical diversity, represented in epithelial tissues by at least 20 different polypeptides. They range in molecular weight between 40kD and 68kD and isoelectric pH between 4.9-7.8. The individual cytokeratin polypeptides are designated 1 to 20. Cytokeratin 1 has the highest molecular weight and the highest isoelectric pH, while cytokeratin 19 has the lowest molecular weight and a low isoelectric pH. The cytokeratins are divided into the type I and type II subgroups, the type II family members comprising the basic to neutral cytokeratins 1-8, while the type I group comprises the acidic cytokeratins 9-20. The cytokeratins are expressed in combinations which characterize the type of epithelium. Cytokeratin 19 is the smallest human cytokeratin (40kD) and is found in many simple and invariably in some non-keratinizing stratified squamous epithelia. All adenocarcinomas tested have been positive for cytokeratin 19.\n\nSource: \nCell culture\n\nApplications: \nSuitable for use in Indirect Immunoperoxidase, Western Blot, Immunohistochemistry (frozen, Paraffin 1:50-1:100). Other applications have not been tested.\n\nRecommended Dilution:\nImmunohistochemistry (Indirect Immunoperoxidase): 1:50. Paraf?n embedded human epithelial tissue sections. Requires protein digestion pre-treatment of paraffin sections e.g. trypsin or pronase.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: Normal breast tissue\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Reacts with the smallest human cytokeratin (Mr 40kD). No crossreactivity with other cytokeratins. Reacts strongly with luminal (secretory) epithelial cells but does not stain the basal (myoepithelial) cells. There is a subset ofsecretory epithelial cells which were Keratin 19 and are thought to be precursors to keratin 19+ cells.
