Apoptosis is mediated by death domain containing adapter molecules and a caspase family of proteases. Certain serine/threonine protein kinases, such as RIP and DP kinase, are mediators of apoptosis. DP kinase (DAPK) is pro-apoptotic calcium-regulated serine/threonine kinase containing death domain. Ectopic expression of DPK induces cell death and suppresses oncogenic transformation. DPK mediates IFNg induced apoptosis. A novel DP kinase-related protein was recently identified and designated DPK2 and DRP-1. Ectopicly expressed DPK2 induced apoptosis in various types of cells. DPK has high sequence homology to ZIP kinase and DRAK1/2, and they represent a novel family of serine/threonine kinases, which mediates apoptosis through their catalytic activities. The messenger RNA of DPK2 is expressed in multiple human tissues. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-1ug/ml Immunohistochemistry (paraffin): 2ug/ml Optimal dilutions to be determined by the researcher. Positive Control: A431 whole cell lysate Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
