DARPP 32, His-Tag, Recombinant, Human (Dopamine and cAMP-regulated Phosphoprotein, Protein phosphatase 1 regulatory subunit 1B, PP1R1B)

DARPP-32 (dopamine and cyclic AMP regulated phosphoprotein, relative MR 32kD) is a cytosolic protein highly enriched in mediumsized spiny neurons of the neostriatum (1). Dopamine stimulates phosphorylation of DARPP-32 through D1 receptors and activation of PKA. PKA phosphorylation of DARPP-32 at threonine 34 converts it into an inhibitor of protein phosphatase 1 (1). DARPP-32 is converted into an inhibitor of PKA when phosphorylated at threonine 75 by cyclin-dependent kinase 5 (cdk5) (2). Mice containing a targeted deletion of the DARPP-32 gene exhibit an altered biochemical, electrophysiological and behavioral phenotype (3). DARPP-32 is a bifunctional signal transduction molecule that controls serine/threonine kinase and serine/threonine phosphatase activity depending on the phosphorylation state of the protein (2).\n\nApplication:\nSuitable for use in Phosphatase assays. \nOther applications not tested.\n\nRecommended Dilution:\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
