Desmocollin 1 (DSC1, Cadherin Family Member 1, CDHF1, Desmosomal Glycoprotein 2/3, DG2/DG3)

Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. The desmosome is subdivided into two regions. The plaque region lies adjacent to the plasma, and is believed to contain molecules that attach the intermediate filament cytoskeleton to the desmosome. The core region is composed of transmembrane glycoproteins that are thought to mediate cell-cell adhesion. Desmogleins and desmocollins are the main desmosomal transmembrane proteins. These desmosomal \nglycoproteins belong to the members of the cadherin family of adhesion molecules. Three different isoforms of both desmogleins and desmocollins have been identified, named as Desmoglein1-3 and Desmocollin1-3. Desmosomal cadherins showed differentiation -specific expression in the human epidermis, although the functional significance of this differential expression is not fully understood. Desmocollin 1 can be found in the upper layers.\n\nApplications: \nSuitable for use in Immunohistochemistry and Western Blot. Other applications not tested.\n\nRecommended Dilutions: \nImmunohistochemistry (Frozen, Paraffin, HEIR): 1:10 in PBS, pH 7.4; Incubation Time 1 hr at RT or overnight at 4 degrees C. \nNote: For better resolution on frozen sections preincubation (directly after fixation) with 0.05-0.2% Triton X-100, for 5-10 min, depending on tissue type, is recommended (2). Without detergent pretreatment a cytoplasmic component might crossreact (e.g. in fibroblasts).\n(Paraffin: slight crossreactivity with a lymphocyte subpopulation is abolished by mild trypsinization after the microwave step.)\nOptimal dilutions to be determined by the researcher.\n\nLocalization: \nLocalizes Dsc1 present in suprabasal layers of interfollicular epidermis; specific cell layers (e.g. in hair follicle root sheath); Hassall bodies (spheroidal formations of densely packed reticulum-derived cells in the thymus)\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH20. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes human Desmocollin 1. Positive on primary keratinocytes, however restricted to local pilles of differentiated cells. All simple epithelia are negative. Reacts with 111kD (form \"a\") and 103kD (form \"b\") polypeptides (Desmocollin 1 splice forms, both at pI 5.2) from human epidermal desmosomes. Species Crossreactivity: Negative with bovine and rodent.
