Samples and standards are added to a 96-well microtiter plate coatedwith capture antibodies, followed by secondary antibody DHEA-S -peroxidase conjugate. The binding reaction is initiated by the addition of a polyclonal antibody specific to DHEA-S. The plate is incubated for 2 hour, washed, and substrate added. The substrate reacts with the bound DHEA-S-peroxidase conjugate to generate a color change. The color is measured at 450 nm wavelength.
DHEA-S is secreted by the adrenal cortex and is a precursor in testosterone and estrogen biosynthesis. The role of DHEA-S is not well understood, however it is primarily produced in the adrenal glands, it is auseful biomarker for adrenal function.
