DJ-1 (PARK7, Parkinson Disease (Autosomal Recessive, Early Onset) 7, FLJ27376, Oncogene DJ1)

DJ-1 protein has 189 amino acids and was cloned from a HeLa cell cDNA library as a novel oncogene which weakly transformed NIH3T3 cells. Transformation by DJ-1 was enhanced by cotransfection with H-Ras or Myc. The expression of DJ-1 is induced by growth signals and it is translocated from the cytoplasm to the nucleus during the S phase, although no nuclear localization signals are present. It has been suggested that DJ-1 is a mitogen-dependent oncogene product involved in Ras-related signal transduction pathway (1). Northern blotting has shown that DJ-1 is ubiquitously expressed in various human tissues.\n\nApplications: \nSuitable for use in Western Blot and Immunocytochemistry. Other applications not tested. \n\nRecommended Dilution:\nImmunoblot Analysis: 1:1000-1:5000 dilution of this lot detected DJ-1 in RIPA lysates from MCF-7 and LNCaP cells. LNCaP cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-DJ-1 (1:5000 dilution). Proteins were visualized using a goat anti-mouse, IgM secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates DJ-1.\nImmunocytochemistry: Has been reported by an independent laboratory to detect DJ-1 in MCF-7, LNCaP and PC-3 cells that had been permeabilized with cold methanol and fixed with formaldehyde.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
