DNA Marker 100bp DNA Ladder is supplied in gel loading buffer at a total DNA concentration of 0.1ug/ul and can be directly applied on to agarose gel. This DNA Ladder is prepared from specially designed plasmid pME-80 DNA, containing pUC, lambda phage and yeast genome sequences. The DNA Ladder contains the following 10 discrete fragments (in base pairs): 1000, 900, 800, 700, 600, 500*, 400, 300, 200, 100. *Reference band. Concentration: ~0.1mg DNA/ml Storage Buffer: Supplied as a liquid in 10mM Tris-HCl, pH 7.6, 10mM EDTA, 0.005% bromophenol blue, 0.005% xylene cyanol FF, 10% glycerol. L3350, Loading Dye Buffer, 6X, 1x0.1ug/ul (in 1ml) (Included): Supplied as a liquid in 10mM Tris-HCl, pH 7.6, 0.03% bromophenol blue, 0.03% xylene cyanol FF, 60% glycerol, 60mM EDTA Quality Control Assay Data Analysis of 0.5ug (5ul) of the DNA Ladder on agarose gel by ethidium bromide staining generates 10 discrete bands pattern. Recommendations for use: 1. Vortex gently just prior to use. 2. Do not heat before loading. 3. Dilute 1 volume of L3350 with 5 volumes of DNA sample 4. Load same volume of DNA ladder and DNA sample. 5. Apply 1ul of DNA Ladder per 1mm of agarose gel lane width. 6. Apply 1-2ul of the DNA Ladder per 1mm of polyacrylaminde gel lane width. 7. For quantification, adjust the concentration of the sample to equalize it approximately with the amount of DNA in the nearest band of the ladder. 8. For DNA band visualization with SYBR Green, GelRed and othe intercalating dyes, do not add the dyes into the sample, use gel staining after electrophoresis or include dyes into aqarose gel to avoid aberrant DNA migration. Storage and Stability: May be stored at 4 degrees C for up to 6 months. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
