DNA PK is the key component of the non-homologous end-joining (NHEJ) pathway of DSB repair in mammalian cells. DNA PK consists of a heterodimeric DNA-binding subunit (Ku70/80) and an approximately 465kD catalytic subunit (DNA PKcs). DNA PKcs is a serine/threonine protein kinase whose activity is greatly stimulated by its recruitment to DNA breaks by the Ku heterodimer Cellular Localization: Nuclear Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional info: This phospho specific polyclonal antibody reacts with phosphorylated pT2609 of human DNA PKcs. Reactivity with non-phosphorylated human DNA PKcs is minimal by ELISA.This antibody recognizes phospho T2609 DNA PKcs as demonstrated: IPed DNA PKcs (using another DNA PKcs antibody) blotted with the pT2609 antibody shows an inducible signal at the correct height after DNA damage. However, unfortunately the antibody picks up an IR-inducible signal in cells lacking DNA-PKcs (MO59J cells) at the same size as DNA-PKcs. We believe that this is 53BP1 which runs at the same size as DNA-PKcs and is also phosphorylated in an IR-dependent manner. Similar results were seen with another phospho DNA-PKcs antibody indicating that this result is general to antibodies to this phospho site, rather than specific to this antibody. This antibody may be a useful marker for DNA damage. It may also still be useful in the study of phospho DNA PKcs T2609. Species Crossreactivity: Crossreacts with Human.
