DNA polymerase comprises an amino-terminal 8kD domain and a carboxyterminal 31kD domain. The N-terminal ssDNA binding domain has a deoxyribose phosphodiesterase activity while the C-terminal domain has a nucleotidyltransferase activity. Mammalian DNA polymerase b, a DNA repair polymerase, is constitutively expressed in cultured cells, but treatment of cells with the DNA-alkylating agents such as N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) or methyl methanesulfonate up-regulates beta-pol level. DNA polymerase b fills single nucleotide gaps in DNA produced by the base excision repair pathway of mammalian cells.
Applications:
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilution:
Immunohistochemistry (Formalin fixed paraffin embedded): 1:100
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
