p56Dok-2 was initially identified as one of several tyrosine phosphorylated proteins derived from bone marrow taken from patients with chronic myeloid leukemia. The protein consists of a predicted N-terminal pleckstrin homology domain as well as several other features of a signaling molecule, including 13 potential tyrosine phosphorylation sites, six PXXP motifs, and the ability to bind to p120RasGAP. It is shown to be 35% identical to p62Dok-1, a recently identified RasGAP binding protein from CML cells. p56Dok-2 is implicated in the regulation of a wide range of physiological responses, including negative regulation of cytokines, enhanced cell migration, filopodia extension and tumor suppression. The adapter protein also plays distinct roles in mediating platelet activation in hemostasis. Human p56Dok-2 gene maps on chromosome 8 at 8p21.3. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:1000 Immunohistochemistry (paraffin): 1:50-1:100 Optimal dilutions to be determined by the researcher. Positive Control: K562 cells Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
