Apoptosis is a normal process involving cell termination, homeostasis, development, and immunoreactivities. Several proteins involve in the process of the initiation of apoptosis, but two main methods of regulation have been identified: targeting mitochondria functionality, or directly transducing the signal via adapter proteins to the apoptotic mechanisms. The TNF-induced pathway and the Fas-Fas ligand-mediated pathway, both involving receptors of the TNF receptor (TNFR) family coupled to extrinsic signals, have been suggested in the direct signal transduction mechanism. In this model, the interaction of death factors such as FasL and TRAIL with the Fas and TNFR results in the formation of the death- inducing signaling complex (DISC), which contains the FADD, caspase-8 and caspase-10, initiating the process of programmed cell death. DR4 (death receptor 4 or TRAIL-R1) and DR5 (TRAIL-R2) are two signaling receptors of the TNF receptor (TNFR) gene superfamily that bind TRAIL. DR4/DR5 are transmembrane receptor proteins containing an intracellular death domain (DD).
