Dishevelled-3 is one of the three human genes (DVL1, DVL2 and DVL3) encoding proteins homologous to Drosophila dishevelled gene, Dsh. Dsh is a member of the segment polarity gene family in Drosophila which plays an important role in the early developmental patterning processes. Dsh is also required for the specification of cell fate and polarity by secreted Wnt proteins, mediating Wnt signaling. Dishevelled-3 mediates Wnt signaling through three key domains: the DIX domain at the amino terminus, the PDZ domain in the central region, and the DEP domain near the carboxyl terminus. Each of these domains likely mediates protein-protein interactions that allow the Dvl proteins to act as scaffolding proteins during Wnt signaling. Dishevelled-3 is widely expressed in human cells and has a widespread role in signal transduction. Dishevelled-3 mRNA is often detected in normal human breast tissues and tumor. Human Dishevelled-3 gene is localized in the chromosomal region 3q27.
