E, Xenopus/Amphibian,portin-2 (CAS; Cellular apoptosis susceptibility protein; E, Xenopus/Amphibian,portin-2; E, Xenopus/Amphibian,p2; Importin-alpha re-e, Xenopus/Amphibian,porter; Chromosome segregation 1-like protein; CSE1L; CAS; , Xenopus/Amphibian,PO2) mediates importin-alpha re-e, Xenopus/Amphibian,port from the nucleus to the cytoplasm after import substrates (cargos) have been released into the nucleoplasm. In the nucleus, E, Xenopus/Amphibian,protin-2 binds cooperatively to importin-alpha and to the GTPase Ran in its active GTP-bound form. Docking of this trimeric comple, Xenopus/Amphibian, to the nuclear pore comple, Xenopus/Amphibian, (NPC) is mediated through binding to nucleoporins. Upon transit of a nuclear e, Xenopus/Amphibian,port comple, Xenopus/Amphibian, into the cytoplasm, disassembling of the comple, Xenopus/Amphibian, and hydrolysis of Ran-GTP to Ran-GDP (induced by RANBP1 and RANGAP1, respectively) causes release of importin-alpha from the e, Xenopus/Amphibian,port receptor. E, Xenopus/Amphibian,portin-2 then returns to the nuclear compartment and mediates another round of transport. The directionality of nuclear e, Xenopus/Amphibian,port is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus.
